I made a presurizing device to check for leaks in my turbo plumbing, and applied lots of silicone to seal the shraeder valve on the plug. If I let it sit for a day, do I still run the risk of hurting the O2 sensor? Should I takes it apart and re-build it without the silicone?

If it does not remain in the intake system for long, you're fine. The silicone becomes a contaminant when ingested and sent through the exhaust.
