Is a 2 inch exhaust setup for a tuned B20 (2130cc ,twin Webers,fast road cam, big valves, 4/1 header,...) enough or should I go for 2 1/4 or even 2 1/2 inch ?

Bigger IS better here! Go for 2.5".
